Comparative proteomics reveals proteins impacted by nitrogen deprivation in wild-type and high lipid-accumulating mutant strains of Tisochrysis lutea ArchiMer

Understanding microalgal lipid accumulation under nitrogen starvation is of major interest for biomass feedstock, food and biofuel production. Using a domesticated oleaginous algae Tisochrysis lutea, we performed the first comparative proteomic analysis on the wild type strain and a selected lipid over-accumulating mutant. 2-DE analysis was made on these strains cultured in two metabolic conditions, with and without nitrogen deprivation, which revealed significant differences in proteomes according to both strain and nitrogen availability. Cultivation of Nannochloropsis sp. in brackish groundwater supplemented with municipal wastewater as a nutrient source BABT

The aim of this work was to study growth potential of the green microalgae Nannochloropsis sp. using brackish groundwater from a well in the semi-arid northeast region of Brazil as culture medium. The medium was supplemented with (%) 19.4, 22.0, 44.0 and 50.0% of municipal wastewater after UASB treatment as a low-cost nutrient source. The results showed that the culture tested was capable of growing in the brackish groundwater even at salinity levels as low as 2 ppt. Furthermore it was shown that municipal wastewater could be used as a sole nutrient source for Nannochloropsis sp.

Culture of Microalgae with Ultrafiltered Seawater: A Feasibility Study ArchiMer

The culture of microalgae is important for the production and maintenance of bivalves. One of the major challenges is to maintain the reliability of microalgae forages over the long term. The aim of this work is to use Ultrafiltered (UF) seawater to cultivate them. Thus, cultures in a volume of 300 L of 2 species of microalgae Tetraselmis and T-isochrysis, were monitored in UF water (membrane pore size: 20 nm) and in sea water usually used on the Ifremer mollusk experimental platform of Bouin (France) (Prefiltration, 3 filtrations and 2 UV). Pesticides used in viticulture create a potential risk for the aquatic environment due to drift during application, runoff and soil leaching. The toxicity of sixteen pesticides and one metabolite were evaluated on the growth of two marine microalgae, Tisochrysis lutea and Skeletonema marinoi, in 96-h exposure assays conducted in microplates. For each substance, concentrations of stock solutions were analytically measured and abiotic assays were performed to evaluate the chemical stability of pesticides in microplates. The artificial medium, in this case named diatom artificial medium (DAM), was designed to maintain long-term cultures of Haslea ostrearia and 19 other planktonic microalgae, and to allow physiological studies related to metal metabolism. This study represents a taxonomic survey of class Bacillariophyceae diatoms found in samples collected in the Iguaçu River (Iguaçu National Park, municipality of Foz do Iguaçu, in the state of Paraná, Brazil) between August 2007 and July 2008. Two sampling stations were utilized, the first located upstream of Iguaçu Falls (Cais Bananeiras Station) and second downstream of the falls (Cais Macuco Safari Station). The study resulted in the identification of 86 taxa, including 74 at the specific level and seven taxonomic varieties. Diet influence on egg production of the copepod Acartia tonsa (Dana, 1896) Anais da ABC (AABC)

Egg production in the copepod Acartia tonsa was evaluated using different densities of the microalgae Thalassiosira weissflogii, Chaetoceros muelleri and Isochrysis galbana. Male and female were kept under controlled conditions (salinity 30, 20°C, photoperiod 12L:12D), acclimated to the experimental conditions and left over a period of 24 h to allow copulation. Algal densities tested were equivalent in biovolume and corresponded to 0, 2.5, 5, 10, 20, 40 and 60.10³ cells.mL-1 of T weissflogii. Diets of tadpoles from a temporary pond in southeastern Brazil (Amphibia, Anura) Zoologia

The diet of tadpoles of 13 anuran species was determined to verify whether food resource partitioning occurs and whether the degree of diet similarity is related to taxonomic affinity. Tadpoles of all species studied were mainly herbivorous, except for these of Leptodactylus fuscus (Schneider, 1799) which were mycophagous. Although some species had exclusive items in their diet, most tadpole species ingested the same items, but differed in the amount of each item consumed. Two guilds were found: tadpoles that feed on diatoms on the pond bottom, and tadpoles that feed on Oedogonium Link, 1820 algae in midwater. Direct and fast detection of Alexandrium minutum algae by using high frequency microbalance ArchiMer

In this paper, a simple detection of a toxic algae, Alexandrium minutum, was developed using highly sensitive quartz crystal microbalance. In terms of performance, compared with other conventional analytical tools, the main interest of our immunosensor is based on a fast and direct detection of these living cells. This system requires the use of one monoclonal antibody directed against the surface antigen of A. minutum. We demonstrate that the whole living and motile algae are caught and detected. The high specificity of the biosensor is also demonstrated by testing several other dinoflagellate species. The frequency shift is correlated to the A. minutum cells concentration. This simple system is potentially promising for environmental monitoring purposes.

Dynamics of the periodically forced light-limited Droop model ArchiMer

The periodically forced light-limited Droop model represents microalgae growth under co-limitation by light and a single substrate, accounting for periodic fluctuations of factors such as light and temperature. In this paper, we describe the global dynamics of this model, considering general monotone growth and uptake rate functions. Our main result gives necessary and sufficient conditions for the existence of a positive periodic solution (i.e. a periodic solution characterized by the presence of microalgae) which is globally attractive. The effect of indigo dye effluent on the freshwater microalgaScenedesmus quadricauda ABU12 was investigated under controlled laboratory conditions. The microalga was exposed to different concentrations of the effluent obtained by diluting the dye effluent from 100 to 175 times in bold basal medium (BBM). The growth rate of the microalga decreased as indigo dye effluent concentration increased (p <0.05). The EC50 was found to be 166 dilution factor of the effluent. Chlorophyll a, cell density and dry weight production as biomarkers were negatively affected by high indigo dye effluent concentration, their levels were higher at low effluent concentrations (p <0.05).
